Academic Achievements
The UltraSpec Lab team has used AI to explore the advantages of supramolecular macrocycles as active drug molecules and novel drug excipients, and has designed and developed a series of supramolecular drug substrates as well as nanomedicine substrates and bionic delivery systems based on supramolecular assemblies. The research results have been published in Nature, Materials Today, Biomaterials Science and many other world famous academic journals.
UltraSpec Lab is currently collaborating with Mathematics of Information Technology and Complex Systems (Mitacs) and the University of Victoria on projects in biomedical diagnostics, substance metabolism simulation, and artificial intelligence drug design.

Sirtuins anti-aging pathway
Sirtuins are proteins responsible for cellular health, also known as "longevity proteins" because they slow down the aging process.Sirtuins are important in regulating cellular homeostasis. However, they can only be activated in the presence of NAD+ and only by enhancing NAD+ in the right way.

By reducing the expression of mTOR, the lifespan of many species is extended. mTOR is a biological pathway that controls cell growth, proliferation, survival and metabolism. When TOR signaling is increased, it triggers many harmful events associated with cancer.

Supramolecular nanodrugs selectively treat cancer by responding to reactive oxygen species and glutathione sequential response to reactive oxygen species and glutathione to selectively treat cancer
The results have been published in the Royal Society of Chemistry's Biomaterials Science.
Cancer cells are usually immersed in oxidative stress at high intracellular reduction levels. Therefore, for specific cancer therapies there is a great need for nanocarriers with sequential response to oxidizing and reducing agents, matching the high oxidizing potential in the tumor tissue microenvironment and the high reducing potential within the cancer cells. In this paper, we report a supramolecular nanodrug consisting of reduction-responsive nanoparticles (NPs) nuclei whose surfaces are modified by oxidation-responsive polyethylene glycol (PEG) derivatives through strong host-object interactions. By this refined design, polyethylene glycolization of NPs not only reduces immunogenicity and prolongs systemic circulation, but also enables oxidative reactive depolymerization in tumor tissues and subsequent release of intracellular payload in response to glutathione (CSH) within tumor cells. As a proof of concept, this supramolecular nanodrug showed specific chemotherapeutic effects against cancer in vitro and in vivo with a favorable safety profile.
This is a supramolecular nanodrug that consists of a GSH-responsive nanoparticle core that is encapsulated by a ROS-responsive PEG shell through a strong host-guest interaction that responds sequentially to the high oxidative microenvironment in tumor tissue and the high reduction potential inside cancer cells.The ROS-responsive depolymerization of PPA-CSNP ensures effective tumor accumulation and intracellular uptake is enhanced, while the CSH response leads to specific payload release within cancer cells, which synergistically produces generally high anticancer activity and low systemic toxicity in vitro and in vivo. Thus, such biocompatible drug delivery systems with precisely designed sequential responsiveness could provide a promising platform for cancer therapy with a favorable safety profile. In addition, supramolecular hosts on the core surface of NPs provide a platform that allows modular functionalization of NPs with other stimuli-responsive shells for a variety of biomedical applications.
